It depends on how far off the strip you wanna go. If you are just going to stay close on the strip or jump over to the Rio I think a cab would be the best route to go. If there are a lot of off strip sites you want to see you may want to rent a car.

kronk is correct. The rio hotel/casino buffet is great. However, might I suggest NOT going during lunch or dinner hours. EVERY 300 lb tub of goo within a 50 mile radius shows up to eat there.
As for nightclubs, RAIN at the palms, "the BEACH" ON paradise ave, and bikiniz nightclub at the rio is pretty good. Here is the second suggestion: GO EARLY if you are going out to a club. get there RIGHT when the line opens up. IF you don't do this, you'll be standing in a line till 2 in the morning or later, IF you get in at all.
wednesdays are hot at club RA at Luxor hotel, mondays (i think) are at studio 54 are good, and there are a ton of other good ones.
some excellent restaurants off the strip are on spring mtn rd, in china town. Some excellent thai, chinese, and japanese restaurants. The nudey bar in china town is sleazy, but the whiskey is cheap, and the women will shove their vagina in your face. Jaguars is right off spring mtn rd on Procyn street. $15 to get in and $10 min for a tiny little drink.

Avoid Circus Circus like it was the plague. Mandalay bay and the Belagio are really nice though. The Monte Carlo isn't bad either. The gym in the Tropicana is pretty complete. It should get you through the week.

Gold's East is the best gym I've been to there. It's about 5 miles East of the Strip on Flamingo (never gets too crowded either). Gold's West on Sahara is OK too, it's more of a new, upkept facility while East is older and more hardcore. The LVAC mentioned is good in that Chicks join free, so it's LOADED with HOT women. Too distracting, I worked out there once (and ended up going to Gold's later that day). I used to live in Vegas, and as far as food/buffets, there are TONS of places to get good stuff, I don't think any one place is head and shoulders above the others. For unreal scenery hang out at The Palms or Hard Rock. Although at the Hard Rock you're kind of limited in what you can do if you're not staying there (e.g. check out the pool. 
) Palms is pretty laid back about letting anybody in to do anything. Fun town, miss it.
