That's kind of an unanswerable question, to be honest. It's typically an instinct that you either have or....you're puddlemonkey. It's actually EXTREMELY easy to pick up on. If you think you can define it by creating a checklist, you're off the mark. It's a smile, a hand gesture, it's in her eyes, it's the way she laughs, it's how she glances at you from the corner of her eye.
Ok now do you think most men have this skill to tell or see. Or are more guys as clueless as above?
Here's an idea for you. This guy that you want...tell him directly.

Not sure, to be honest. I'd say most guys I've chilled with do, but then again, we tend to surround ourselves with likeminded ppl. I've also seen plenty of totally clueless dudes.
So, in short, there's probably a pretty healthy mix of both in the world. Up to you to figure out which type of guy you're dealing with and adjust your game accordingly.

LOL i am the same way. until they cross the line verbally or physically i just never know. I am really good at flirting and just wait for girls to get so fed up with my teasing that they make the first move. ive had sex with a girl before and even the next day im like idk if she likes me and im all shy lol. wtf.

If we seem like we can't tell its because you must be one of the girls we think are way to pretty or have too nice a body to possibly want to be with us, so we...yeah we can't tell.

I used to be the most oblivious retard when it came to understanding when a girl was flirting with me, and thus I came off as being really stuck up and picky, when that wasn't the case at all. I'd be talking to a girl, would leave, and then a friend with me would be like, "Dude, she wants you," and I'd be like, "Nah, she's just putting up with small talk."
Looking back on it, this was probably the main reason why I didn't have many girlfriends in high school. My inability to pick up on female interest led me to not really talking to girls much, which made me come across as a too-good-for-everybody type.
Now, I can pick up on it almost immediately. I was able to do this by really examining all aspects of a particular interaction. Like somebody else said, there's not really a formula or checklist, but it's just her mannerisms around you, the way she laughs at you compared to the way she laughs at other people, if she compliments you for things that aren't impressive at all. Definitely the arm touch or the "shoulder nudge" thing (idk what to call it), and ESPECIALLY if you are saying intentionally retarded things and she pretends to find them humorous, when you KNOW her sense of humor is completely at odds with what you just said. I swear, I could say something like, "Boy the Holocaust sure was awesome, wasn't it? and 50% of girls who are into me would pretend to laugh. An extreme (and absurd) example, of course, but you get the point.
I can even tell when a girl is trying to pull me in just from online talk. I met this girl the other week. First, she friend requests me. That could be a 50/50 (I don't think I'm God's gift to women, so I don't automatically think the slightest acquaintance indicates romance). Then, she just out of the blue IMs me, and just says "Hi." I respond with, "What do you want?" If a girl wasn't interested, she would probably just never respond again, because that's a pretty rude response, but she writes, "Nothin, LOL." I decide to really be a douchebag, and write, "OK, are you an 8 year old retard or do you need something?" "I just wanted to say hi, jeez LOL."
OK, well that's pretty obvious. I was a dick, and she rolled with it. Not every girl who IMs me is into me at all, it's a completely different feel. It's hard to explain. You just know it when it happens.

Now he is either A) not interested or B) trying to play it real cool and not act too interested in this super hott chick that wants to see him again. Being too subtle is not a good thing. I'm not talking about coming on like gangbusters, because that's sure to chase most normal guys off...but a single follow-up is not a bad idea. For example, simple poke if he's on your Facebook. Or a simple hello on MSN...even better if you do the MSN thing when you know he won't be online...that way it's not forced conversation, just a nice hint that you would like to hear from him again.
If you get nothing back after doing that, he's not interested. Move on.

But you know the worse thing by far is having the dense, but otherwise really nice, guy that you're totally not digging, but he keeps coming around and calling. Oh damn there was this one guy....so sweet....but so not interesting or sexy at all, that just didn't seem to get that I was trying to get rid of him nicely and diplomatically. Dude that is the worst.

I'm pretty friendly so everybody I met seems to take a liking to me. Girls laugh at my jokes and like to hang around me, but I never interpret that as "interest" because. . . You know I don't want to come off like a douche who makes a move on a chick just cause she is friendly with me. Nothing worse than making a move on a chica who just wants to be friendly.
I may have probably missed out on some girls that were interested in me because I can be so clueless about it some times. Unless a girl is noticeably flirtatious, I just never tried to move in...
That may be the reason why I'm the only guy who has been on EF this (9yrs?) long and who hasn't gotten with any chick here lol - I may be an Admin, but the regular members here get more action than I ever have(or will).

KB pretty much said it though, it's mostly body language. A lot of small, non-verbal signals that speak louder than words. If someone is good at picking up on that they can tell, if not, well they should learn/study it then.

they're paying attention to what I'm saying. looking at my eyes while talking, not being distracted by friends, seem to be at ease, laugh/smile alot during the miriad of stories I tell, touch my hand and lean forward when they're talking..... stuff like that. If guys are more into themselves, they'll miss alot of that. Age takes care of most of the BS though.
FYI, If the "I'd like to see you again" thing didn't get thr response you were looking for, maybe he's just not making himself clear yet, or he's shy, or he a socially inept sumbitch.
