Spliting Up Quads and Hamstrings

I've always done legs as a muscle group/day just like chest, back, etc.
Does it make sense to focus on quads by themselves just how one would maybe focus on biceps separately from triceps?
Also, besides stiff dead lifts, leg curls and seated leg curl machine, what are some other good hamstring isolation excercises?

What are your goals? Are you BBing or PLing, maybe something else?

Either way, I am a big proponent of exercises that mimic life - even if you are eBBing. You body was designed to work is a certain way (like squatting down and picking something up). I also try to avoid machine work as much as possible with the thinking that the machine is telling you how to work rather than your body. That said machines do have their place.

Have you tried a glute ham raise? Or a reverse hyper?

How much are you doing with deadlifts and squats?

Romanian Deadlift and GHR's tear my hamstrings up, especially RDL's in the 6-8 rep range and GHR in the 12-15 rep range, I find that works well. Differant variations of legs curls you could use ie lying dumbell leg curl, standing cable leg curl, seated machine leg curl, lying machine or cable leg curl or reverse hypers as bblazer said although Iv never tryed any of these, only things I have ever done for hams are RDL, GHR and lying leg curl. RDL I have found to be my favourite. You could also try good mornings but I always find these hit the lower back pretty hard, more so than hams.

I split them.. one day- hams, abs, calves...another day- quads & abs.

for hams, i also do good mornings, reverse hack squats & barbell LUNGES! here some others for ya-
