Sounds like a preamp tube to me, probably at the front end. Try tapping on them with a wooden pencil and see if one is more microphonic than the others.

Yes, when the amp is on. You'll be listening for any unusual noises through the speaker when you tap the tubes with the pencil. If they have metal shields on the, you'll need to remove those first.
