To eat:
Vinegar Hill House (You have to get the Red Wattle pork chop)
Peter Luger Steakhouse (IF you can get a reservation)
ANY food off a cart on a street corner! Pretzels and Kinish's especially!
Cap off the evening with a carriage ride on Central Park, and then head to any number of coffeehouses, live music bars, or blues joints in Greenwich Village.
Just some of my favorites. Being from Upstate New York though (about 3 hrs. northwest of NYC), I've gotten to do things a million times, and can basically do them whenever I want. You'd have to prioritize by your interests if you only have a limited amount of time.
