The half-life of arimidex is around 46 hours.

It will reach full blood hormone levels around 5-6 days after you start using it when dosing 0.5mg EOD.

So I've been using arimadex for one week and my water retention is still there. I'm only shooting 250 mg of test e per week and taking 50 ngs of winny per day. Should I increase my Adex to 0.5 Ed?

No don't increase to that. At 250mg test per day, you are probably at too high of an arimedx dose anyways at 0.5mg eod. What does your diet look like? That is where 99% of people get water retention.

Diet is very good. Not used to having the water retention blow me up ten lbs in five weeks.
