It is up to you bro. Generally, you don't want to use them to train with, but rather as an "aid" on heavy sets only. When I used them(I havn't squatted heavy in a long time), I would usually go up t0 500 for a set of 3, then back down for one or two burnout sets. Progressing up to the 500 pound set, I would go 135, 225, 315, then 405. I wouldn't apply the wraps until the set of 405. Reason for that is two-fold. One, because you want to train as many sets without the wraps, so that the tendons get properly warmed up, as well as having done as many sets as possible without the training aid, so that you know that the muscles are the reason those particular pundages are being moved. The second reason I applied the wraps at 405, is because you want at least one set using the wraps to be completed before you use them on your heavy set(s). I wrap my knee wraps TIGHT bro. I stetch them out as far as possible, and have someone pull as hard as they can on the end of the wrap after I am finished, in order to get them pulled as tight as possible. Being that they are so tight, I always liked to have one set before my heavy(again) to warm up to that amount of squeezing. It can feel like the veins in your calves are ready to explode when you wrap that tight, at that amount of weight. I always need on semi-heavy set, wrapped tightly, to get aclimated.
After the heavy set of five, assuming I would do just one, I would come down for a set of 365, then 225. Those were done without wraps.
