chemistry question

First, find someone willing to lend you a cell stock of hGH-transformed E coli.. I don't think you would find it cost or time efficient to construct your own expression vector and clones.

Then, once you have that done, it's all down hill from there....

Are you starting your own biotech company or trying to be the worlds first UG source for HGH?

well for my undergrad research in microbiology, im attempting to isolate, purify, sequence completely and mass produce (well on a scale to be studied furhther) a protein called the ISS protein which is been shown to have some correlation with immunity from a strain of pathogenic ecoli (avian). people have been almost successful but end up with something like several extra bases on each side of the extracted protein. its continuation of the thesis of the prof that taught me general micro.

working off the top of my head, its pflag or pmac or something like that is what im workin off of.

our goal is to at least have some idea to design an antibody for it, or at least have a decent crystal of the protein.
 
i just really want to try it and see if its feasable in an undergrad lab... ive never actually isolated and purified a protein, just isolated and identified an "unknown" and ive done some PCR from genetics and something a little bit in my first biochem class.

It really isn't.

Granted, you could find an infinite number of ref's or good protocols for synthesis of recombinant hGH (because, next to insulin, it is probably THE highest produced protein).

You will end up needing equipment (or access to equipment) that will be far more expensive considering the reward you will get.

75% of the work would be: using specially designed primers to PCR, purify, ligate into expression vector, then transform comp bacteria cells.

Even if you went with my suggestion of finding someone who already has stocks of these cells prepared, you would still be left with the task of growing up the cells and harvesting, purifing hGH... It will be daunting.. I promise.

Hands on knowlege is what you will need.. (assuming you have access to a biochemistry lab at a university).
