The x16 slot can move 16 times the amount of data that a x1 can move, therefore it is your best slot for a video card.

I'd use that as your primary video card slot for your center / main monitor. Put another card (cheaper) in your x4 slot that has two monitor ports to drive the two side monitors or ancillary monitors...

That's what I'd do....
